									<p><span lang="EN-US"><b>New Alzheimer&#8217;s genes</b></span></p><p><span lang="EN-US">An international research team led by the Alzheimer Center and the Human Genetics<br />Department of Amsterdam UMC has discovered two new &#8216;Alzheimer genes&#8217;.<br />Colleagues Henne Holstege and Marc Hulsman have made a significant contribution<br />to this research.</span></p><p><span lang="EN-US">Evidence has also been provided for another new gene. A hereditary damage to one of<br />these genes can lead to a significantly increased risk of Alzheimer&#8217;s disease. The study is published in Nature Genetics.</span></p><p> </p><p><span lang="EN-US">The study compared more than 32,000 genomes from Alzheimer&#8217;s disease patients and healthy individuals. The researchers looked for rare harmful genetic mutations with an<br />increased risk of Alzheimer&#8217;s disease. They found that deleterious mutations in the genes ATP8B4 and ABCA 1 can lead to an increased risk of Alzheimer&#8217;s disease. This has not been observed before. In addition, the researchers discovered that deleterious mutations in the ADAM10 gene probably also lead to an increased risk of Alzheimer&#8217;s disease. However, additional confirmatory research is needed for this gene. Together with ADAM10, there are now nine genes of which it is known that hereditary damage to the gene can lead to an<br />increased risk of Alzheimer&#8217;s.</span></p><p><span lang="EN-US"><b>Insight</b></span></p><p><span lang="EN-US">All genes found are involved in maintaining brain health. For example, the two newly discovered Alzheimer&#8217;s genes are involved in the processing of the Alzheimer&#8217;s protein amyloid and in the function of the brain&#8217;s immune system. The finding that precisely these genes are more often disrupted in Alzheimer&#8217;s patients provides more insight into the processes that go wrong in the brains of people with Alzheimer&#8217;s disease.</span></p><p><span lang="EN-US"><b>Susceptibility to Alzheimer&#8217;s</b></span></p><p><span lang="EN-US">The younger the Alzheimer&#8217;s patient, the greater the genetic nature of the disease: it is estimated that more than 90% of the risk of Alzheimer&#8217;s disease in people under the age of 65 is due to hereditary factors, while the risk of Alzheimer&#8217;s disease around age 75 for about 60-80% can be explained by hereditary factors. However, with the genes that have now been found, the risk-increasing hereditary damage can only be determined in a minority of these &#8216;young&#8217; Alzheimer&#8217;s patients. That is why the researchers are now comparing much larger groups of Alzheimer&#8217;s patients with healthy individuals so that they can identify more new genes that are involved in a higher susceptibility to developing Alzheimer&#8217;s. This is slowly but surely revealing which genes influence susceptibility to Alzheimer&#8217;s.</span></p><p><span lang="EN-US"><b>Start on time</b></span></p><p><span lang="EN-US">With these new developments, it will become increasingly possible to estimate the degree of susceptibility and the nature of the susceptibility to this disease per person, long before symptoms manifest themselves. In the future, personalized treatment can then be started in time, before damage occurs in the brain. During this meeting he presented his new book :Morgen gaat het beter&#8221;. </p><p>Henne Holstege had the honor of receiving the first copy from Humberto Tan himself. In his book, a hunderd centenarians are visited and interviewed. All centenarians were asked the same questions. &#8220;At what point in your life were you the happiest? Have you known fear in your life? Do you still have any dreams? And do you have a health recommendation?&#8221; </p><p>The 100-plus team is very proud of Henne. We are thrilled that she received the first copy.</p><p> </p></div>								</div>

<p><strong>Biochemist Dr. Henne Holstege of Amsterdam UMC receives a BrightFocus Alzheimer Research Award with a grant of € 250.000, intended for scientific research. The aim of her current research is to learn from healthy centenarians how it is possible that their brains continue to function well into a very old age. Holstege investigates the influence of genetics, the immune system and the structure of Alzheimer’s proteins in the brains of these special people. The grant enables her to expand the 100-plus project with research into the presence of Alzheimer’s proteins (blood biomarkers) in 500 bloodsamples from centenarians, their children and children-in-law. To this end, she is entering a collaboration with Prof. Charlotte Teunissen, head of the Neurochemical Laboratory at Amsterdam UMC. </strong></p>

									<p>How is it possible that some people suffer from the symptoms of dementia at a young age, while others live beyond 100 years of age with no signs of cognitive decline? To find out more about this, Holstege set up the 100-plus study in 2013. She s project leader f this research and heads the Genomics of Neurodegenerative Diseases and Aging section at the Department of Human Genetics of the Amsterdam UMC.</p>

<p><span style="color: #6ab650;"><strong>100-plus Study<br /></strong></span>It is known that reaching very old ages occurs within families. This indicates that genes are strongly involved. We know that some hereditary defects cause or increase the risk of dementia. Holstege and her team are also looking at the genetic abnormalities in centenarians that prevent someone from developing dementia.</p>
<p>The question is: how do the genes of the centenarians do that? One way to learn more about the protective mechanisms of the centenarians is to examine the characteristics of Alzheimer’s disease in these centenarians. Which characteristics do they have or do they not have? To this end, Holstege and her team carefully examine the brain functions of the centenarians, their genes, the composition of their blood and even their stools. About a third of the centenarians participating in the study agree to brain donation after death for the purpose of this study. This, together with all the other data collected, is a gold mine for the researchers because they can learn which properties in this special group contribute to the maintenance of their brain functions.</p>

<p><span style="color: #6ab650;"><strong>Blood-biomarkers<br /></strong></span>Alzheimer’s proteins build up in the brains of patients with Azlheimer’s and – to a lesser extent – also during normal aging. These proteins are known to be detectable in the blood as well. Previous research indicates that they mirror the Alzheimer’s processes that take place in the brain; we call these substances blood biomarkers. That is why the researchers would like to know to what extent Alzheimer’s proteins are also present in the blood of the centenarians and how these relate to the functioning of the brain.</p>

<p>To investigate this, Holstege has entered into a partnership with Prof. Charlotte Teunissen. She is an expert in developing and measuring blood biomarkers for neurodegenerative disorders, including Alzheimer’s.</p>
<p>Simply taking a tube of blood can detect Alzheimer’s processes in the brain, possibly even before there are clinical symptoms. In the future, this could contribute significantly to the timely administration of medication that counteracts Alzheimer’s disease.</p>
